Learning outcomes This course "Theoretical and Hands-on Course: NMR for metabolic profiling" provides an introduction to the study of metabolism using Nuclear Magnetic Resonance (NMR) technique, from the preparation of biological samples to the analysis and interpretation of the obtained data. It includes theoretical lectures, in an online format, and practical in-person sessions at the NMR laboratory of the Health Sciences Research Center at the University of Beira Interior (CICS-UBI).

By the end of the course, students should be able to:
(i) understand the basics of NMR spectroscopy and its application in the study of metabolic processes;
(ii) understand the main metabolic pathways;
(iii) prepare biological samples for metabolic analysis by NMR;
(iv) acquire NMR spectra from different biological samples for subsequent cellular metabolism analysis;
(v) analyze and interpret the data from the obtained NMR spectra.
Syllabus 1. Theoretical part:
1.1. Introduction to NMR: principles and applications
1.2. Unveiling metabolism using NMR – applications overview
1.3. Main Metabolic Pathways on carbon Metabolism accounting for biosynthesis and bioenergetics
1.4. Ovarian Cancer: cues for metabolic adaptation, early diagnosis, and follow-up
1.5. Overview of NMR metabolomics in disease research and drug discovery
1.6. Metabolic Fluxes by 13C NMR isotopomer analysis
1.7. Expanding the coverage of the metabolome using 15N and 31P NMR
1.8. Metabolic studies using deuterated water and 2H NMR analysis of metabolites enrichements

2. Practical part:
2.1. Incubation of cells with 13C-labeled substrates, using glycolysis inhibitors or hypoxia inducers
2.2. NMR analysis of experimental media collected before and after applying experimental conditions
2.3. Preparation of cell extracts and metabolic analysis by NMR
2.4. Discussion of the results obtained.
